Rebecca – or Revy as she prefers others to call herself – is not the type of person anyone would want to cross. A member of the Lagoon Company, she is the one Dutch and the others turn to if they want someone dead really fast (there are rumors she committed several unsolved murders in New York City). She has an almost-passionate zeal for killing, and is often seen grinning maniacally during a shoot-out. Dutch once commented that Revy's personality was a little closer to Charles Whitman, the University of Texas sniper who went on a shooting rampage after murdering his mother in 1965.

Revy earned the nickname "Two-Hand" because of her ability to shoot with accurately with a gun in each hand... namely her customized chrome-plated Berettas. She also has a knack of being able to avoid bullets that are fired at her... traits the Chinese American picked up while surviving the mean streets of New York City before she made her way to Roanapur. She is also well-versed in other firearms, which range from grenade launchers to heavy sniper rifles.

Spike Spiegel is a tall and lean 27-year-old bounty hunter born on Mars. The inspiration for Spike is found in martial artist Bruce Lee who uses the martial arts style of Jeet Kune Do as depicted in Session 8, "Waltz For Venus". He has fluffy, dark green hair (which is inspired by Yusaku Matsuda's) and reddish brown eyes, one of which is artificial and lighter than the other. He is usually dressed in a blue leisure suit, with a yellow shirt and Lupin III inspired boots. A flashback in Session 6 revealed it was his fully functioning right eye which was surgically replaced by the cybernetic one (although Spike himself may not have conscious recollection of the procedure since he claims to have lost his natural eye in an "accident"). One theory is that his natural eye may have been lost during the pre-series massacre in which he supposedly "died". The purpose of this cybernetic eye is never explicitly stated, though it apparently gives him exceptional hand-eye coordination - particularly with firearms (Spike's gun of choice is a Jericho 941, as seen throughout the series). In the first episode, when facing a bounty-head using Red Eye, Spike mocks him, calling his moves "too slow". At first, this seems like posturing on Spike's part, but even with his senses and reflexes accelerated to superhuman levels by the drug, the bounty cannot even touch Spike. A recurring device throughout the entire show is a closeup on Spike's fully-natural left eye before dissolving to a flashback of his life as part of the syndicate. As said by Spike himself in the last episode, his right eye "only sees the present" and his left eye "only sees the past." Spike often has a bent cigarette between his lips, sometimes despite rain or "No Smoking" signs.

As Trigun unfolds, it becomes apparent that Knives believes that humans are parasites that need to be exterminated. He wants to create a peaceful and fair world with Vash and his other plant brethren, and he believes the only way to do this is by killing off all humans. Knives never claims to be a god; simply a superior being to homosapiens. If Knives were able to kill all humans and begin creating his "Eden" on Gunsmoke, it is very likely that he would act similarly to Vash, and act as a kind and loving individual. This can be seen in the final episode of the anime, where Knives peacefully rests amidst the oasis he has created and greets Vash as his brother arrives, but this could actually be a facade of placidity.

Uvogin is a typical Enhancer: determined, head-strong, simple and sometimes disrespectful. He is barbaric in manner, viewing even pain merely as a nuisance. Like all members of the Phantom Troupe, Uvogin never hesitates to kill and is remorseless in doing so. He particularly enjoyed to fight against skilled opponents, but typically rushes into battle without a plan. Although he prefers to fight alone, Uvogin becomes stronger when fighting with a partner because he fights better when there is someone else to protect. Uvogin is best friends with Nobunaga Hazama and the two bring out the best in each other during combat. Uvogin, like all of the other members of the Troupe, is absolutely loyal to Chrollo and the Spiders' ideals. He dislikes carrying money and claims he has absolutely no use for it as he firmly stands in his belief that one should steal whatever they desire.

Jonah (ヨナ Yona?), real name Johnathan Mal (ジョナサン・マル Jonasan Maru?) is a former child soldier and the newest and youngest member of Koko Hekmatyar's squad.Jonah tends to be quiet and rarely smiles, and it is unusual for him to laugh. His parents were killed in an attack on his village by its enemies who were armed with weapons sold by Kasper Hekmatyar, Koko's older brother, and he initially wanted revenge for this as well as for the treatment that he received from Kasper before he was recruited. Jonah's recruitment was not completely voluntary, as he exchanged his service in return for Kasper aiding three young orphans that he cares about..
